I have keywords with two phase, one ranking on in 100 while second one is ranking 500+.

Keywords is look like php programmer and php programmers, difference is only “s” at end of keywords.

php programmer - 100
php programmers – 500

Which keyword, I have to target for seo, so I can get my both keywords ranking on search engine.

as i know you have to target "php programmer" keywords with proper backlink.

It totally depends upon your requirement which keyword you want to target.The keyword you will target will go high in rankings.

You should target both "php programmer" as well as "php programmers" because both are searched in good quantity.

The keyword for which your page is describing the relevant content is the best keyword for keywords ranking in the search engines.
